Daisy Jones grows up rich but largely unattended in Los Angeles, finding companionship and danger in the clubs of the Sunset Strip while still a teenager. Singer Simone Jackson becomes her closest friend and helps her understand the compromises demanded of women in the industry. In Pittsburgh, brothers Billy and Graham Dunne form a band that gradually becomes the Six. Billy emerges as its forceful leader and songwriter; Graham plays lead guitar, Eddie rhythm guitar, Pete bass, Warren drums, and Karen Sirko joins on keyboards. Billy's girlfriend Camila believes in the group and moves to California with them. The album sessions strengthen private attachments elsewhere: Graham and Karen begin a secret relationship, although Karen insists that it not alter her place in the band. Eddie's frustration grows as Billy changes parts and dominates decisions. Aurora becomes a major success and transforms Daisy Jones & The Six into one of the country's biggest bands. Publicity emphasizes Daisy and Billy as a pair, intensifying feelings that are already difficult for them to contain. Daisy briefly leaves the pressure behind and impulsively marries Niccolo Argento, known as Nicky. His charm initially feels liberating, but he encourages rather than checks her dependence on drugs. On the road, Billy remains sober and tries to keep distance between himself and Daisy, while Daisy interprets his restraint as rejection. Graham and Karen continue their hidden relationship; Graham imagines it becoming permanent, while Karen values both him and the independence her career provides. Teddy suffers a serious health crisis but returns to work. As the band's success expands, its public image appears unified, while its members are carrying incompatible desires and increasingly fragile private arrangements.

The Aurora tour magnifies every unresolved conflict. Daisy's drug use becomes severe, and Nicky abandons her during a medical emergency rather than seeking help. Simone intervenes, and Daisy finally orders Nicky out of her life, though sobriety remains uncertain. Billy feels both drawn to Daisy and responsible for keeping the band functioning, while maintaining the boundaries that protect his marriage and recovery. Camila understands more of their bond than either admits. Karen learns she is pregnant. Graham is pleased by the possibility of a child and marriage, but Karen knows that motherhood is not the future she wants and fears surrendering the career and autonomy she fought to obtain. Eddie's resentment of Billy's control also reaches a breaking point. By the time the band arrives in Chicago, its success can no longer conceal that Daisy and Billy, Karen and Graham, and the musicians as a collective are approaching decisions that cannot be reversed.

In Chicago, Camila tells Daisy to leave Billy and begin saving herself, then reminds Billy of the life and family he chose. Daisy sees that her hope for Billy is helping keep both of them trapped and decides to go. After an emotionally charged final performance, she leaves the band and eventually becomes sober. Karen has an abortion because she does not want a child; the difference between her future and Graham's ends their relationship. Billy refuses a return to drugs and goes home to Camila. The remaining tour is canceled, and Daisy Jones & The Six never perform together again. In later life, the members describe separate careers and families: Daisy adopts children, Billy stays sober and devoted to Camila, and the others continue along their own paths. The interviewer compiling their conflicting memories is revealed as Julia, Billy and Camila's daughter. Camila has recently died, and Julia includes a message in which her mother encourages Billy to contact Daisy when he is ready, offering the former collaborators a possible new conversation after decades apart.

At the Chicago show, Camila separately confronts Daisy and Billy about the feelings they refuse to resolve. Daisy recognizes that remaining near Billy will destroy the life he has chosen and that her own marriage and drug use are endangering her. She performs with the band one last time, then leaves. Karen ends her pregnancy and her relationship with Graham because she does not want the domestic future he desires. The group cancels the rest of the tour and never plays together again. Daisy becomes sober, adopts children, and builds a quieter life; Billy keeps his promise to Camila, remains sober, and raises their daughters with her until her death. The others continue in and around music on separate paths. The unnamed interviewer is revealed as Billy and Camila's daughter Julia, who has assembled the history after her mother's death. Camila's final message encourages Billy to contact Daisy if he is ready, leaving open the possibility that the two surviving collaborators may speak again without undoing the family life Billy and Camila shared.
