During the French and Indian War, Major Duncan Heyward escorts Colonel Munro's daughters, Cora and Alice, toward besieged Fort William Henry. They reach the fort, only to endure its surrender to Montcalm and the massacre of the departing British by France's Native allies. Magua abducts the sisters, driven by revenge against Munro and a desire to make Cora his wife. Hawkeye's group follows through Huron and Delaware territory. Disguises and escapes free Alice, while Uncas is revealed before the Delawares as the heir of a revered Mohican line and leads a pursuit of Magua. Cora refuses Magua and is killed by a Huron; Uncas dies trying to save her. Hawkeye shoots the fleeing Magua. Alice survives, but the joint funeral of Cora and Uncas ends the rescue in grief, and Chingachgook mourns his son as the last of the Mohicans.

In 1757, as French and British forces contest the American frontier, Major Duncan Heyward leaves Fort Edward to escort Colonel Munro's daughters Cora and Alice to their father at Fort William Henry. They travel with the singing teacher David Gamut and a Huron guide named Magua. The guide leads them away from the main column along an uncertain route. They encounter the frontier scout Hawkeye and his Mohican companions, Chingachgook and Chingachgook's son Uncas. The woodsmen identify signs that Magua has deceived the travelers. When challenged, he escapes, and Hawkeye's party assumes responsibility for bringing the vulnerable group through territory where hostile warriors are already close.

Hawkeye guides the party to a concealed refuge among caves and rocks by the falls. Huron pursuers discover them, and a prolonged attack exhausts the defenders' ammunition. Hawkeye, Chingachgook, and Uncas leave by the river so that they may survive to seek help, while Heyward remains with Cora, Alice, and David. Magua captures the group and reveals his grievance: after Colonel Munro punished him for drunkenness and expelled him, he resolved to take revenge through Munro's daughters. He demands that Cora become his wife. She refuses. Before the captives can be killed or carried away, Hawkeye and the Mohicans return, ambush the Hurons, and free them.

The reunited party presses toward Fort William Henry, moving cautiously through French lines and using darkness, water, and captured passwords to avoid detection. They finally enter the besieged British fort and reunite Cora and Alice with Colonel Munro. The defenses have been battered, and General Webb at Fort Edward refuses the reinforcement Munro needs. Heyward declares his love for Alice after first creating confusion by speaking to Munro without naming which daughter he means. French commander Montcalm reveals Webb's intercepted message and offers honorable surrender terms. With resistance no longer defensible, Munro accepts, and the British prepare to leave under a promise of protection.

The surrendered garrison and its families march away from Fort William Henry, but the French-allied warriors, angered by the terms and eager for plunder, attack the defenseless column. Montcalm does not stop the massacre. Magua uses the chaos to seize Cora and Alice again, with David carried along as a harmless captive. Heyward, Munro, Hawkeye, Chingachgook, and Uncas survive. After the killing ends, they search the devastated road, discover signs that the sisters live, and begin tracking Magua's party northward. The military mission has become a personal pursuit across country controlled by Huron and Delaware communities.

The rescuers follow a trail that Magua has tried to conceal and eventually reach the vicinity of a Huron village. David, allowed to move because the Hurons consider him harmless, tells them that Alice is held nearby while Cora has been placed among neighboring Delawares. Uncas is captured while scouting and condemned by the Hurons. Heyward enters the village disguised as a French healer, while Hawkeye uses a bearskin disguise. Their improvised deceptions allow them to reach Alice, free her, and escape with help from Chingachgook. Hawkeye then exchanges identities with David so the singer can leave while he remains positioned to aid Uncas.

Uncas escapes the Huron village, and the companions turn to the Delaware settlement where Cora is held. Magua arrives to assert his claim over the captives and expose his enemies. Before the aged leader Tamenund, Uncas reveals the tortoise emblem that identifies him as descended from the Delawares' honored ancestral line. The community recognizes his rank and rejects Magua's power over most of the prisoners. Custom nevertheless allows Magua to depart with Cora, who remains under his immediate control. Uncas rallies the Delaware warriors and leads the rescuers after him toward the Huron refuge in the cliffs.

The Delawares and their allies defeat the Huron defenders and pursue Magua into the caves and cliffs. Cora again refuses to submit to him. One of Magua's companions kills her before Magua can complete his threat; Uncas reaches them moments later and is killed while trying to save her. Magua kills Cora's murderer but is forced toward the edge of the rocks. Hawkeye shoots him during his attempted escape. Alice is safely restored to Heyward and Munro, but the victory is overwhelmed by the deaths of Cora and Uncas. At the funeral, the Delawares mourn both and honor Uncas as the last young bearer of his line. Chingachgook grieves that he is now the last of the Mohicans, while Hawkeye affirms the friendship that remains between them.

The Delawares recognize Uncas as the heir of an honored Mohican line and rally behind him, but their pursuit cannot save Cora. At the cliffside refuge, Cora refuses Magua; another Huron kills her, and Uncas is killed trying to reach her. Magua in turn kills that warrior and attempts to escape, but Hawkeye shoots him as he hangs from the rocks. Alice is rescued and reunited with Heyward and her father. The Delawares hold funeral rites for Cora and Uncas, mourning them together and imagining a bond beyond death, while Munro grieves and Hawkeye remains beside Chingachgook. With his son gone, Chingachgook calls himself alone, the last of the Mohicans. Hawkeye answers that friendship still joins them, but the close offers consolation rather than restoration: the rescue succeeds only in part, the younger Mohican line has ended, and the surviving families leave marked by the violence of the frontier war.
